Matchups

Anivia - A difficult laning opponent, her egg makes it so you cannot kill her early due to mana/skill rotation issues. If you are able to kill her with a jungle gank, abuse the lead and win the lane, always take note when her egg is back up. She is able to push the lane and roam easily forcing you to stay in lane.

Cassiopia - Destroys you, especially in lane, her harass has a much longer range than you and is able to out trade you if she applies poison. Do not get caught facing her ult when she is level 6. Seek the opportunity to fight her while her ultimate is down.

Fiddle (The champ we all hate) -

Whether it be mid or jungle, Fiddlesticks is an *******. Mercury Treads are a must against him, you cannot fight him in a 1v1 match-up, unless some how you are able to get a lead. Although, late game does get easier if you can survive before hand. If laning against Fiddlesticks, try and seek to trade with him, if he is coming out on top, focus more on farming until your jungler is able to aid you. Wards are your best friend against Fiddlesticks, especially level 6+.

Ori - A very tough match-up. Since your snare does not cancel their ability to cast spells, she is able to auto attack you and cast spells when you try to approach her even within your prison. She is tough, but with all match-ups it does not mean you cannot outplay her. Look to dodge her ball whenever possible and watch for her ultimate at level 6. Her auto attacks are very strong due to her passive.

Xerath (The champ that kicks ***) - Xerath does very well against Ryze, even if he goes into his Siege mode. As you try to approach him he will try to land his stun combo off, harass him pre-6 and dodge his spells. Upon level 6 he can kill you if he is able to stun you, if possible dodge his stun/ult combo, if done, that is your window to kill him.

Optimal

Baron/Dragon/Buff: Q E Q W Q R Q
Bursting an Enemy: Q W E Q R Q

(Although, with the item reworks in the past, as well as the new masteries, a majority of the time you no longer have Max CDR as Ryze which does affect the way your combo spell order is. At max level with full CDR you would be able to Q at the beginning and in between every single other spell, but without that CDR there is a slight delay).

Tip: While pushing a lane or taking wraiths, don't hold onto your ultimate as it has an extremely low cooldown, especially with your Passive. And while many do not use this or do not know this, spells that are still traveling to the target can apply your ultimate AOE if activated before it reaches the target. This allows you to Q -> R -> Q with full AOE utilizing your passive to his fullest.

If you take Teleport instead of Ignite as one of your masteries, be sure to let your team mates know before hand that you want to gank their lane. Especially bot lane, have them ward the 2nd bush in lane behind the enemy, once the enemy has pushed, it should secure 2 kills.
