Introduction
Welcome to my guide to playing Vayne, the Night Hunter. This is my first (submitted) guide, so be gentle. Things such as item build and runes will get moved around as I get more feedback, both from playing as her and from the comments here. Don't be afraid to leave a message!
Pros:
-Excellent harasser
-Great assassin
-Great anti-tank character
-Great zoning and denial
-Great escapes
-Sweet Shades, bro.
-Plays like Batman
Cons:
-Squishy when focused (Then again, who isn't)
-Not a great farmer
-Can't tumble through walls or small ledges
- Really needs to keep on target or lose her damage output. Attacking another target resets her Silver.

(Innate): This ability makes her an exceptional chaser, combined with tumble, no one will ever get away from you.
: This skills kinda your bread and butter, you want this, and you want to spam this.
: I like to call this skill, "Caitlyn: Abridged." Basically, pick a target, stick to that target and you will destroy that target.
: Okay, I love this ability. Not only have I gotten some great kills with this, but this ability has save my life more times then I can count because of that knock back.
: Standard steroid to make you even deadlier.

Masteries:
21/0/9 is fairly straightforward for a ranged carry.
Runes:
Greater Mark of DesolationArmor penetration marks or go home.
For quintessences I recommend Greater Quintessence of Desolationarmor penetration, but you can use flat health quintessences for an early game boost.
For Seals and Glyphs you have a bit of flexibility here and it all comes down to preference. You can either go defensive and use flat armor or Greater Seal of VitalityHP/level seals with MR/level glyphs. Or, you can use MP5/level seals and cooldown per level glyphs. Personally, I'm rolling with MP5/level and cooldown/level.

Work in Progress, things will get tossed around here. I'm still working on picking more situational/optional items.
For starting items I recommended either getting as a starting item, it gives you some great early game survivability and damage. Or you can get /3x if you'd rather just go straight into the build.
For early game your core consists of:
This, in my opinion, gives big bro Vayne a good mix of damage, mobility, and survivability early game.
For boots take either or . My preference is for Berserker's for even more attack speed. And now that they added , you have another option for CC heavy teams over merc treads.
Black Cleaver: Honestly, this item is perfect for Vayne. You get attack speed, damage, and a stacking armor debuff, what's not to like?
Trinity Force: This item, is amazing on Vayne. Sheen synergizes so well with tumble, Phage ensures that targets won't escape and zeal helps you destroy enemies. This item rules.
Infinity Edge: This item is awesome. It just is. Combined with your fast autoattacks and relatively high crit chance, this item adds some great damage.
Banshee's Veil: The go-to defensive item against casters. However, if the enemy's a bit more more AD heavy, you might wanna skip this.
Bloodrazor: The anti-tank weapon. Combined with your you'll tear tanks apart with this.
Other Items
Bloodthirsters: Oh how much I love this item. I'm not sure this item works well with Vayne, I feel like she could use more damage, and normally to get more damage you get one of these. However, stacking up a bloodthirster on Vayne is a bit of an issue with her lack of aoe.
Cloak and Dagger: CC heavy team? Grab this.
