Introduction - PLEASE READ THROUGH ALL THE GUIDE
Hi, this is my second guide on one of my favorite champions.
As we all know, Vayne is a great carry, ranged BUT very squishy so be sure to work as a team to get those early kills that will give you the most money. Vayne doesn't have trouble getting kills early game, either Solo or laning.
I hope you enjoy this guide and don't rage at me about it. It has worked wonders for me so feel free to comment and vote :)

Pros / Cons
Well these are some of the things you have to consider when playing Vayne:
She is a sexy Night Huntress ;)
She is an early killer.
Sick movement speed later on so, great stalker.
Tumble helps her run and stalk.
If you take care not to die your attack damage will be HUGE with Sword of the Occult.
She can help gank due to her huge movement speed to go through lanes.
Condemn is a very good escape route either pinning them to a wall or just a little behind
Silver Bolts is a killer, trust me.
Final Hour makes the world go round :P
She is squishy as hell so don't get caught between too many minions or you will regret it.
Tumble doesn't go through walls.
You will be targeted first on team fights. (Most likely)
You will almost always be asked to go mid so be prepared.
She has no AoE :( so almost no farming for her until mid game.
This guide is only if you are a good player and won't die much because most of the damage comes from Sword of the Occult at early game
Well, as I have said before. This guide is not how a Vayne should be played, just 1 of the best ways I have used her and it has worked wonders. So please try it before you vote.
This will give us that extra damage boost at the start and open path to our early game Sword of the Occult.
Since you will be left with a little gold and we are not starting with Doran's Blade, this will be a good item to get. You will be left off with 25 gold so if u think u can wait around 7 seconds more or less after minions spawn to get a second one then do so.
Sword of the Occult
Since you most likely are gonna be owning with Vayne you will get a lot of kills and assists. (WARNING, don't blindly follow into turrets. Just make the kills you are able to) If you go steady without dying (Which is pretty easy if you are cautious), you can have around 10 stacks for mid game by now, if not more.
Boots of Swiftness
Pretty good, easy and obvious choice. She is a stalker, she needs to actually reach the champions she is following. Also, these are cheap boots.
This item is wonderful in Vayne. This makes those hits with Tumble WAY more significant, and of course it will help you play Tumble more often due to its CDR (Cooldown Reduction) Also, let's not forget that it has Armor Penetration. People will be so afrsid of you by this early game that they will pee their pants.
This item is your next choice. Will give you a little extra damage, still keep the CDR and boost your Armor Penetration. AND it's active goes perfectly with Vayne, more Movement Speed and Attack Speed? Why, yes, thank you very much.
Well, this is the time when you realize you have had to get potions all the time because of your squishiness. Well not to worry, due to your most likely huge damage with SotO by now you will gain enough HP with this item and of course it leads us to our next big item.
This item works wonders on Vayne, she will deal huge damage and heal a part of it, what is better about it is that it charges itself with even more damage and Life Steal for each kill (Yeah, minion kills too) so it is a great asset at this point of the game.
The Black Cleaver
This MUST be your next item, it is simply amazing for Vayne. A lot of attack damage. A lot of attack speed. Oh and what's that? It ALSO reduces the target's armor? Sounds good to me, I'll buy it! Remember the more hits you get the more times you will trigger Silver Bolts.
Well, if the match has taken so long to get to THIS point AND you have lost most of your stacks on Sword of the Occult you can Aim to get this one. You can either sell Youmuu's or the SotO if u feel like the game is about to end and you have no stacks left anyway.
OTHER ITEM OPTIONS:
If you are a Fan of Silver Bolts then this will do the job for ya. Also if you aim for a mid game Infinity Edge this will help even more, And of course the 15% movement speed will make no one run from you and you will get to team fights faster if you die or had to recall.
Awesome item with Vayne, and in this build can work wonders. If you are all on the aggro side and don't wanna spend a lot on The Bloodthirster then buy this instead. With the damage and armor penetration you have on this build a well put Sheen on a third attack for Silver Bolts using Tumble can get you A LOT of kills.
I think I heard someone saying that this also works on the Silver Bolts damage. So if you are an attack speed guy/gal and use Condemn a lot then go for this. What to change for it? The Black Cleaver if you like.
Well this is my choice on Runes and I hope you like it. Of course you are welcome to choose whichever runes you want.
Greater Quintessence of DesolationGreater Quintessence of Desolation
An Awesome Rune for what we need. Armor Penetration, and with 3 of these is a rough 9.99% Armor Penetration right at the start of a game.
Greater Seal of Fortitude
This Rune will give you the little extra health at the start to help on your squishiness.
Greater Mark of Strength
Well this is what will make your life easier early game. Early kills mean more money and more stacks for SotO
Greater Glyph of Strength
Well I think it's obvious why we go 21/0/9. She is a heavy DD, and she wants levels and money fast so we use that to our advantage on the utility part. I think there is not enough to say in here although you may try 21/9/0 if you think you are too squishy and are not too used to playing Vayne. That way you may not be killed by minions that often :P
And then again, I choose this because Vayne is a stalker so she enjoys both slowing opponents and making herself faster
Helps with her Squishiness. She won't die so easily using this. Also, it slows them down allowing you to kill them or just stalk a little at least and make them return to base.
As I have said before, I prefer this over Flash on Stalker characters. If I flash, I will just appear beside them and they will run again. If I use ghost I will hit them sometimes by still following them.
You may not want to max your certain skills right away but just keep alternating them. Trust me is the best way.
Vayne ruthlessly hunts evil-doers. She gains 40 flat movement speed when moving towards an enemy champion.
Vayne tumbles, maneuvering to carefully place her next shot. She rolls toward the cursor and her next attack deals bonus damage.
Well, is Vayne's Signature Move. Awesome boost on attack. GREAT ability overall. us this as much as you can, Vayne NEVER runs out of mana lol.
Cooldown: 6 / 5 / 4 / 3 / 2 seconds
Bonus Damage: 40 / 45 / 50 / 55 / 60 %
Vayne tips her bolts with a rare metal, toxic to evil things. The third consecutive attack or ability against the same target will deal additional damage plus a percentage of the target's maximum health as bonus true damage. (Damage against monsters is capped at 200.
What can I say? One of the best passive abilities in the game dealing True Damage :)
Base True Damage: 20 / 30 / 40 / 50 / 60
Max Health to Damage Ratio: 4 / 5 / 6 / 7 / 8 %
Vayne draws a heavy crossbow from her back, and fires a huge bolt at her target, dealing physical damage and knocking them back. If they collide with terrain, they are impaled, dealing bonus physical damage and stunning them for 1.5 seconds.
Believe it or not I am not a fan of the attack. BUT for the people who are this is a great asset to Vayne. Pin them to a wall by carefully placing your tumble then they will be most likely dead by the time they spent stunned.
Cooldown: 20 / 18 / 16 / 14 / 12 seconds
Initial Physical Damage: 45 / 80 / 115 / 150 / 185 (+0.5 per bonus attack damage)
Collision Physical Damage: 45 / 80 / 115 / 150 / 185 (+0.5 per bonus attack damage)
Readying herself for an epic confrontation, Vayne increases her combat skills for a few seconds. During this time, Vayne gains increased attack damage, 1.5 second stealth after using Tumble, and triple the bonus movement speed from Night Hunter.
I deeply enjoy this ultimate above most ultimates on other champions. It will give you kills 90% of the time.
Bonus Attack Damage: 25 / 40 / 55
Duration: 8 / 10 / 12 seconds
Early Game is simple:
Soloing: Be careful, get kills when you have to. Don't chase into turrets. Get rid of the minions if there are too much or you will die by them instead of an enemy champion.
Duoing: Your partner must know the importance of you getting kills so let them start fights and you finish them, remember not to follow too close to turrets as you will most likely die of 2 turret kills at Early Game.
Mid Game is simple as well:
Soloing: If you just killed someone go and help lanes in trouble to gank and get more kills or assists and will avoid your team mates from gettign killed. After that go back into your lane to avoid unfriendly pushes.
Duoing: Same as above, go help gank mid 2v1 to ensure kills.
Late Game is not as easy but it shouldn't be hard:
If you are winning: Rush into the enemy's base and help your team mates. Be careful of ganks while you are at it. And careful of champions closing in to your base, get rid of them first.
If you are losing: Since you will be fast enough switch from place to place defending your turrets killing the most squishy champions first. Then once you are safe make ALL your team push mid.
Well I hope you have liked my second guide as I put a lot of effort into them xD
Again i say this is not a guide of how YOU SHOULD play her, is just a guide that I made of how I love to play her. Of course I always play her in a lot of different ways. Like going full attack speed with a little damage, or pure critical strike with Infinity Edge :P and stuff like that.
Please, if you are going to down vote give me a good reason and not stupid reasons like I have seen in other guides or as in my other guide. I quote:
Sorry for downvote but AP leona is useless. Did u even play her this way before writing this guide? Her AP ratios are worse than Alistar´s after his nerf. Even magic penetration doesnt help it. She deals much more damage just with trinity force than with your full end game build.
The best way you can play her is to get :

I was building an AP Tank. Not a full Tank. Tanks are boring and it was obvious that person didn't read through my guide. Just looked at the build and voted.
lol AP tank. You do realize her AP scaling is terrible, right?
Again someone that did not read my guide. I posted the reason I go with AP, and to make it short it was to avoid half-baked kills. To ensure kills and not let people go alive going back to base.
