Introduction

This is only how I have found it is best to play Vladimir. Take my advice at your own risk, though that's not to say that it's not good advice.

Runes

The magic pen is meant to give your transfusion a little bit of extra "oomph" early game. The ability power is meant for the same thing. The addition to your health from your quints and glyphs will serve you well throughout the entire game.

Masteries

9/0/21 is pretty typical for a caster. Note the way I skipped the mana masteries.

Items

Doran's shield gives you an extra health (mana and ability power) boost early. The Guise does virtually the same thing. However, it allows you some extra ability power as well. I personally prefer it over other alternatives. Rylai's is one of Vlad's bread and butter albums. The slow is fantastic. Hextech leads into Will of the Ancients, which is fantastic for all casters. Rabaddon's and Void Staff need no explanation.

Skill Sequence

The purpose of the skill sequence is to max out transfusion by 9, this is necessary for causing trouble for your laning opponent. However, the rest of the abilities can be leveled at your leisure. Those are just my preferences.
