For Masteries I go 1-16-13. I find this Mastery Tree to be the most effective, mainly because you are incorporating tankiness with utility which is great for an offtank. The 3 points into Meditation really do make a difference. Early game, your mana usage greatly suffers until you require a Tear of Goddess.
Another choice is is 9-21-0, taking 9 points into the 15% magic penetration. Yorick's Omen of Prestilence and Famine both are magic damage that scale off of AD. I wouldn't recommend this Mastery Tree, but if you don't have a full rune book, or just feeling like being a pro tank, I guess you can take this route.

If you've played with a Yorick, I'm guessing you see many of them level up their Q at level 1. Well I am here to tell you that THAT IS NOT THE WAY TO GO! First of all, your Q only gives you an additional 30 damage at level 1 with about a 10 second cooldown. You need to be up close and personal to the person your fighting which most of the time, won't happen at level 1. This does not do sufficient amount of damage without Sheen or Trinity Force, which is the main reason you should not take it at level 1.
The reasoning for maxing out your Q last? Well, at the early stages of the game, you will not have Trinity Force, which is the item that you need to deal massive amounts of damage from your Q. Your better off with having longer ranged attacks for harrasment during the laning phase. By maxing your Q first, the enemy knows you are vulnurable and can attack you from far. They will not even think of you as a threat.

For spells I take Flash and Exhaust. These are easily exchangable for other spells.
What spells you can take
Ghost: Prefer ghost over flash? well take it instead
Ignite: I can't stand having enemies walk away with less than 50 health. Ignite solves those problems.
Teleport: Since Yorick is a great pusher, teleport is also a great spell for him. You should switch this with exhaust though, since Yorick lacks escaping abilities.
What spells not to take
Clarity: Although Yorick has mana problems early on, you will not need this. Once you require your Manamune, you will not have any mana problems.
Smite: This isn't a jungling build. Yorick can jungle, but is bad at it compared to other champions.
Fortify: Leave this for the real tank.
Heal: You already have an ability for lifesteal, you don't need this.
Clairvoyance: This spell fits perfectly into the support role, leave it for your support.
Revive: lol
Rally: Rally is just bad now...I guess you can use it to help you push more, but in my opinion, that's really not worth it.

Doran's Shield
This is probably one of the best items to start out with as Yorick. Doran's shield gives you great lane sustainability with it's health regeneration. Combine that with your E's lifesteal, and you will be staying in your lane for a long time.
Meki Pendant
From the feedback you guys are telling me, Meki Pendant is another great item to start off with. Since Yorick is so mana starved, this will work great. Plus it will make you get your Manamune even faster.
Manamune
THIS ITEM IS A MUST FOR YORICK. Your passive proves this. Yorick's passive is for every ghoul that is up, your basic attack deals 5% bonus damage. Your ghouls die off VERY quickly. That is why you must spam your abilities as much as you can. Without a Manamune, you cannot do this, making you weaker than you should be. Once your Manamune is fully stacked, you can spam abilities as much as you want.
Mercury's Treads
Boots are just preference. The best boots for Yorick in my opinion are:
Mercury's Treads
Ionian Boots of Lucidity
Boots of Swiftness.
Trinity Force
This item is almost essential for every offtank. Trinity Force has everything Yorick needs. Movement speed, Health, Mana, Crit chance, and attack speed. Trinity Force also procs with your Q, making you deal massive amounts of damage.
Warmog's Armor
Finaly you start to look very beefy after you achieve this item. When your build is complete, warmongs will have a great effect to your attaack damage from atma's impaler.
Sunfire Cape
THIS ITEM IS INTERCHANGABLE
If the enemy team has mainly physical champions, take Sunfire Aegis
If your going against a team with AP attacks, take Banshee's Veil
If your going against a mix of AP/AD champions, then take Guardian Angel
Atma's Impaler
Now that you have Atma's Impaler, you are more tankier, do higher amounts of damage, and have a higher chance of critical hitting. Your Q can crit, which makes this item so good for Yorick.

[Levels 1-6]
Buy your Doran's Shield or Meki Pendant and head off to your lane. If possible, lane with a ranged champion and avoid melee champs. If your laning with another melee champion, you will be harrassed forever and will be forced to recall sooner than you thought.
At these levels, the most important thing to do is FARM. When your health is low, use your E on the squishiest champion to harrass them and to heal the max from your ghoul. Nearing level 6, you should have enough money to atleast buy Manamune or be very close to it.
[Levels 6-13]
In these levels, you should have completed your Manamune and your boots of choice. Trinity Force should be started at around these levels. Since you have decent AD and mana now, you can harrass like a boss now. Make sure you are using your abilities to get mana from your Manamune. If you are in a fight, USE YOUR ULT ON YOURSELF RIGHT WHEN THE FIGHT HAPPENS. Your revenant deals 50%-75% of your AD (depending if your level 6 or 11) which is HUGE. You can probably win any 1v1 situation if you have your Manamune and Sheen by level 11 with the help of your ultimate.
Levels 13-18
As you are approaching late game, you will notice Yorick gets very strong at these stages. Once your Trinity Force and Warmog's Armor is complete, your pretty much unstoppable.
If a team fight approaches, MAKE SURE YOU USE YOUR ULT ON YOUR AD CARRY. This is essential. By level 16, your ultimate's revenant deals 100% of the chosen ally's damage. This has to be one of the best ultimate in the game. Instead of having 1 AD carry in a fight, you now have 2. It makes such a big difference and will help the team win teamfights.

Have you learned an interesting thing about Yorick and willing to share it with us? Well post below so I can add it in this section!
1. Denying Rammus
Is Rammus using his Powerball to catch up to you for the kill? Well don't even worry about that anymore! Use your W on an area that rammus WILL go to. He will crash into your ghoul, stopping the animation from his Powerball, giving you a quick get away.
