"Hello, I'm Binglederry. Here's my guide on twitch. I know some people will disagree with my setup, but I'm sure someone will get something out of it. Making it all worth while.

Unlike his lesser evolved cousins, Twitch (as he calls himself) can talk and read and interact
with the world as well as any of the other sentient beings in Valoran. Twitch has come to the League of Legends to represent himself, the only one of his kind, on the Fields of Justice.
While Twitch's true motivations remain known only to him, his prowess in the arena is a matter of public record. As a creature of the sewers, he is able to blend in with the shadows and ambush unwary victims of his choosing. He has collected a significant quantity of the toxic soup that collects in Zaun's underground, and he coats his crossbow with it. The effect of being shot with one of Twitch's crossbow bolts is extremely debilitating. Even without his poison, he will outright infect those he chooses to attack, as is the nature of a plague rat. As an act of desperation, Twitch may rapidly unleash a flurry of bolts at his foes. He madly peppers a focused area, though with mercifully reduced damage.

Runes: Some people prefer a mix of armor penetration, critical chance, and physical damage runes, but with the item build I go; I'll have plenty of damage and crit. I personally find critical damage runes to be the best for twitch/shaco/gangplank.
1) Now that rally was nerfed twice, I perfer exhaust and flash or ignite.
(Passive) - Deadly Venom -
Twitch's attacks infect the target with Deadly Venom, which deal damage each second, stacks up to 6 times, and lasts up to 8 seconds.
Q - Ambush -
Twitch hides in the shadows, waiting for a nearby enemy. If he attacks, he will gain bonus attack speed. After 1.25 seconds, Twitch becomes stealthed for 20 / 30 / 40 / 50 / 60 seconds. If Twitch attacks a unit while Stealthed, he gains 20 / 30 / 40 / 50 / 60 attack speed for up to 10 seconds, depending on how long he was stealthed before attacking.
W - Debilitating Poison -
Infects a target with a crippling poison, slowing his movement speed.
Slows nearby enemy champions' move speed by 20%, plus 10% per Deadly Venom they have.
3 / 3 / 4 / 5 / 5 second duration.
E - Expunge -
All nearby enemies secrete Twitch's toxic venoms from their bodies, dealing damage for each stack.
Deals 30 / 60 / 90 / 120 / 150 (+1 per ability power) magic damage to each nearby poisoned enemy, plus 20 / 30 / 40 / 50 / 60 additional damage per stack of Deadly Venom applied to them.
R - Spray and Pray - (Ultimate)
Twitch closes his eyes and fires his crossbow as fast as he can, spraying piercing arrows ahead of him.
Twitch gains 40 / 80 / 120% attack speed and his attacks become 1000 range line missiles for 7 seconds, dealing 100% damage and poisoning all enemies hit.
If your laning solo, you might want to put 1 point into Deb. Poison early on incase someone tries to gank you.

Early Levels (1-8): Focus on last hitting creeps and if you have a good laning partner; gank if the opportunity arises. Keep up the harassment, but make no attempt to tower dive for a kill. Keep in mind your ultimate has a long range, so if they are camping by the tower with low health, you can try for a kill as long as they don't have a stun/fear.
Mid-Game (8-14): By now you should have your malady and boots, and working on your black cleaver. I bounce between lanes to gank if someone is low on health, if not, then stick to farming in your lane until you get your black cleaver.
Late Game (14-18): Depending on how well the early - mid game ganking went, you either just finished your black cleaver or are getting close to another B.F. sword to build into an infinity edge. Once you get your black cleaver, its a whole new game. You'll be doing a large amount of damage from there on. Just keep an eye out for team fights, but never initiate, as you'll get sh*t all over.
I'm only posting the ones that are useful for twitch, the rest are up to you.
Offense - 3/3 Deadliness
2/4 Sorcery or Archmage's Savvy (These two are up to you, you just need the next tier)
3/3 Brute Force
1/1 Improved Rally (Rally took the nerfhammer up the ***, so its up to you on this one)
Laning partners: Blitzcrank is my favorite laning partner, with his rocket-grab -> power fist combo and my dps combined, they're almost guaranteed to die. Especially if one of us has an exhaust up. If you can, lane with someone who has some sort of stun, slow or burst damage instead of lets say... Soraka :P"
