Ok a tank is a char(a champion)who take a lot of dammage, he is one of the most important char in the game, if you don't have a tank the game is dead...
The tank is here for take the carry's dammage and save your carry.
He have 3 important ability:
Life(HP):survive in general
Armor: to reduce AD's dammages
Magic Resist(RM): to reduce AP's dammages
With this build you reduce ~75% AD and ~65% AP and you have many HP.

Cleanse
Teleport
Heal

Revive
Smite
Ignite
Exhaust
Rally
Clairvoyance (let the support take it)

So Mark Armor, Seal MR , Glyph HP, and quinte HP

Actualy is the most equilibrial choose of runes :D

Eventually you can take HP regen in glyph

Not really difficult ;D

(Flash)=>Q=>E=>W=>R ----> the BASIC skill sequence

E=>R ---->To tank/destroy a tower (E give you AD)
