With this initial Rune, Mastery, and item setup, you start out each game with a good amount of HP, 10% spell vamp, 6% life leech, and 10% bonus magic damage.

How can you beat that?

Start off by getting Mark of the Assassin (Q) and take the Red buff lane.

If no one on your team is jungling, you want to kill the golem creeps on your lane. If you do it by yourself, just kill one golem then run away. If you can get an ally to help you, you should be able to kill them both. Either way you get half a bar of XP.

Now run down your lane and see who you're matched up with. If your enemy is aggressive, you might want to pop your health potion right away so you don't get destroyed. Otherwise, use your life leech and spell vamp to get your life back. This can be done quickly by using (Q) on a minion then attacking it right away. This ability also make it very easy to get last hits with Akali.

When you hit level 2, get your Twilight Shroud ability (W). This ability saves my life at least 10 times per game. You should only use it to avoid taking damage from the enemy or to try and keep them away from your tower if they get close. If you are being chased, pop it down before they reach you and possibly stun you. And here's where flash comes in handy. Since most enemies will wait in your circle until it goes away (especially if you have low health) run to an edge of your circle that is furthest from the enemy and right after you get outside of the line pop flash and you're home free...most of the time.

Akali is a master of last hits. Get as many as you can early, get items and start ganking.

Once you get the hextech revolver, you can leave your lane and start ganking away while clearing the jungle along the way.

This is a pure AP build with Akali which gives massive burst damage.

Akali is usually targeted in team fights and that's why I use the Cleanse/Flash combo. Jump in and get a few hard hits in the cleanse (if needed) and flash out. Or you can use your (R) ability to get out if there is an enemy minion near by to jump to.

Stay behind your tank and wait for them to initiate.

In late game, Akali can take down almost anyone (except some tanks) 1v1 with all her spell vamp and life steal so don't be afraid to jump someone that's all alone.

Good luck with Akali, she's a lot of fun to play and destroy with.
