Vayne the night hunter, well, dont think i can say something clever about her introduction or else... But i have some thought. So, ppl i played against or with, most of them think that Vayne is OP, well, dont think so, not coz riot nerf her, just coz, well, i dont see her az OP. i.e. when u play in 3v3 (most played match for me in pub games) and u see loading screen with singed or noc, or some other OP :) i think damn, lose.... again... But not with Vayne, see her and think oh well nothing special... annoying yes, OP, not!

For runes, nothing new here, i use set-up shown above in 'Rune Build", but there is some options, it depends of ur play style. There is some choices:
Seals-
-So u can change atk.speed runes for armor runes (will give u +13 armor), or take crit chance runes, which is also good, but in that case u will be 2 dependent from atk. speed items...
Glyphs-
I dont think u should change it for cdr or smthing else, coz mres is highly needed since she is squishy and Vayne dont receive mres with lvl's... at start of the game u will have 49 with +6 from masteries and +13 from runes. and ur base 30, which is good. But again, u may change it az u like.
Marks-
Well, nothing to comment here, obvious choice, everybody wants it, everybody needs it.... but mb u can change few runes for crit chance if u want to crit more often...
Quints-
Again, obvious choice is armor pen., but again u can change 1 or 2 runes for crit chance, atk speed or some resists armor/magic.

Nothing special here, u can set masteries az u like, just make 21 in offence and make the other 9 az u like, its just my set-up.

Build i like to use is for atk speed, but it seems that a lot of ppl use sheen az a core item, well, i tryed it, with atk speed i like it better, coz silver bolts r just insane.
Boots, zeal, is my core, after that, if everything went smooth, i rush for black cleaver, but if there is a lot of nuckers in enemy team, or i just need some more health i go for m.res or phage and mb even frozen mallet, but its rare. So i just use 2 core items for atk speed, some crit chance and mov. speed, other items r situational and depends on how the game is going. But the build is shown in "Items Purchases" is the one i use often. If there is combined team with mages, ad... and u need some armor, i advise u to take Guardian angel it has what u need, and u will be less focused (yeah, ur the prior focus target).
One last thing, if ur good with active items, always remember to activate, rly good item instead of black cleaver or, even core item is SoD, good vs evasion champs especially jax (nemesis of all the ad) and u gain +30 armor on activation which is great.

Exhaust and flash, popular choice. Again u can go for ghost-flash or exhaust-ghost.
Don't think there is a better choices.

Summary, Summary, Summary, Summary.... Well what can i say, Vayne is great i like the champ, and im even dont like ad much... try it, make ur own build's, he is rly flexible with items, and runes....
Don't rly have anything to say... my first build so i'm kinda confused, had a lot of thought before i start, but now they gone...far. ^^
