Her passive, nighthunter, is the skill which makes her snowball so much harder. Not a single enemy can escape you.
Tumble is a free repositioning spell and in combination with your ultimate, it's a short repositioning spell which makes you invisible! It also enhances your next auto attack, so use if often. In laning, use this to reset your autoattack timer, so you can shoot twice, while the enemy could only shoot once.
Silver bolts is what makes Vayne such a killing machine late game. Every third auto attack you deal a % of the enemies max hp as true damage. Try to get these three hits off the enemy in the laning fase, because it also adds a flat amount of true damage to your attack, which makes the harass much stronger.
Condemn is your escape spell and the spell to set up kills in laning fase. If you get the stun off in laning fase, you are most certainly going to outdamage the enemy (on equal terms). This makes her work so well with other ccing characters, like Alistar, Janna and Taric.
Last but not least, her ultimate, final hour. This gives Vayne stealth on her tumble and provides alot of extra attack damage. It has a very low cooldown, so you can use it often.

For runesI use armor penetration marks, flat armor seals and magic resistance (MR) per lvl glyphs. For quints I use flat AD.

There are certain assassins who make me go slightly more defensive, but with good positioning, you shouldn't be able to be hit 95% of the teamfights. For summoner spells I nearly always use:
-Flash
-Cleanse
Why? Because it makes repositioning much easier teamfights and you are literally uncatchable with stealth+tumble+flash+cleanse.
The only time I don't use cleanse is when the enemy has a knock-up heavy team. You can't cleanse a knock up, so you're better off getting exhaust for that pesky bruiser who jumps on you while you're in mid-air.

There are many exceptions, but once you learn how to play this game at a high level, you will most likely be able to choose your own build, depending on the situation.
Just remember you need full stacks on your bloodthirster to make it worthwhile and infinity edge is the highest damage item in the game. Also keep in mind black cleaver gives flat armor pen and last whisper gives a percentage armor pen.

Make sure you are behind your team, in a position where enemies can be knocked off you by your support. Once you are able to do this and deal damage (not important yet on who you deal the damage, as long as you deal it), you should start working on getting in positions where you can deal damage on the most favorable characters: the squishy ones, preferably the ones dealing damage.
I always prefer supports who can keep me safe, like Alistar and Janna. Being able to blow away the AoE damage deals and the tanky dps makes it already worth having such a support.
It also gives alot of fun in laning, because Alistar can set up wall kills for you and Janna disrupts the enemy so much and gives you so much extra damage, that you will be able to outdamage almost anyone at level 6.
