As I said in the guide, this guide is mostly for Varus players who have quite a bit of mana problems. The manamune covers this very well, especially early game, and I have had pleanty of gamesd where it did fill up fully. Also, do remember that the manamune does give AD based on your maximum mana, so that's a nice bonus, too. Also, this allows for more farming with your skills, in addition to making more harass with your Q.

Why Manamune? You won't fill it up over the course of the entire game, you should only be using your skills to harass when you have 3 stacks of blight / in a team fight, and in both situations you should have kept high enough mana for both..
