The AD glyph just adds 1 extra 1 AD so its stronger early, and I've had good success with that. It's just something I've used ever since Chaox showed to his stream that it is a setup he uses.
I don't really support, so I wouldn't know too much. I play mid lane for my ranked team on cyber gamer, since I moved from top.
I think that it is perhaps a bit too longwinded, but making it succinct as you suggest is kind of counterintuitive given it is designed to help newer players, but I will fix it somehow.
Also, I wouldn't recommend Magic pen quints, even for ryze. I think he need movespeed, else Mana or Health as he is short ranged.
That was the focus of my last, I just didn't add many champions because I forgot I had it there xD

1. The one AD glyph is NOT a good idea. I understand it's for rounding off purposes, but AD glyphs give stupid stats (0.28 as opposed to 0.95 on reds), and furthermore take into account that AD carries have no natural MR growth. So 1 MR > 1 AD (actually 1 MR > 0.28 AD).
2. Your support section is wrong. Gold quints are core. Gold seals are not, especially on aggressive supports.
3. This is just me, but I'd lay out the choices in a way which is a bit more succinct, such as:
Mid Laner (AP) - You can ONLY choose from runes in the list below.
Reds: Magic pen. No others are allowed.
Yellows: Scaling HP, scaling AP, flat mana regen, flat armor
Blues: Scaling AP, flat MR, scaling MR
Quints: Flat AP, movement speed, magic pen (only on champions who don't scale well with AP, such as Ryze and Lulu.
4. The focus on your last chapter should be on champions with unusual rune pages, which you already have. The biggest example is of course Akali. Other examples can possibly be rammus (flat armor reds and yellows), Sona (mana regen yellows) and Soraka (the only champion in the game who uses CDR blues).
In all, I think this is a pretty good job.
