Runes

You can use whatever you want. I recommend hybrid runes. For Nasus in jungle nine Greater Seal of Armor is great. The magic resist is important to because of the magic animals. And the armor penetration for if you wanna carry in a lane. Here is one for sample

Masteries

Nasus is harmable in early game. So the defense is quite important.

Summary

Nasus is a mid-late game champ. But in late game can be very brutal if you can handle him. In PvP not really needed. But when your team defending the nexus you can break in any lane(I recommend top lane.). So if you made right you can easily solo towers until the enemy's nexus.
In teamfights he cant be very effective. You only can use his Wither and Spirit Fire.
