You don't need any defensive runes if u know how to play passive and use wards to deny enemy jungler ganks.

Anivia is pretty easy champ to farm in late game, but really hard to last hit at early game. So little AD is good for your masteries to make sure you hit all of your last hits. At late game you can just use R + Q to clear whole minion wave.

So here are all spells:

Q - Flash Frost: Anivia summons a very slow moving ball of ice that chills and damages everyone in the path. When the ball explodes it stuns anyone in the area. Remember you can always press Q when ball flies trough air to explode it faster.

W - Crystallize: Anivia creates a wall of ice what blocks all movement.

E - Frostbite: Anivia blasts her wings and creates a freezing arrow to air , dealing a medium amount of damage. If the target is slowed by Flash Frost or Glacial Storm , the damage is doubled.

R - Glacial Storm: Anivia summons a driving rain of ice and hail to damage her enemies and slow their advance.

I use AD Masteries 21/0/9 to make sure i don't lose any last hits and it makes tower pushes slightly faster.

Here are champions what are strong against Anivia:

Fizz: Fizz can easily dodge her Q and W with his E and burst her so easily.

Kassadin: Kassadin can dodge her Q and W aswell with his R, and burst anivia down in late game.

LeBlanc: LeBlanc burst Anivia down after lvl 3 so easily. Playing vs LeBlanc just try to farm under your turret and try to hit Q when LeBlanc uses her W to make sure you hit it.

Talon: Talon can dodge Anivia's Q with his E or with R. And Anivia is so weak early that Talon can burst her down before lvl 6.
