Do you mean your keystone mastery? You can get this by selecting one of the bottom masteries.

I am quite new to this game. I finally became LV 30 recently.
When a game starts, it shows everyone's selected summoner spells. Everyone has three spells but I only have two. How do I select the third spell?
I have purchased three runes and saved it. I also have chosen masteries and saved. But I don't think it's now assigned to my champion even though I select them before the game starts. How can I apply runes and masteries?
