This might be a tricky question, but how can I tell if I'm improving or not? LoL does a pretty good job of keeping your W/L around 50/50, so it's not a linear judgement like most games.

41/35 isn't shabby, although I'm 1 and 4 in my last five (I feel comfortable laying that last loss on the team. I was 5-1-3 when the lanes broke down and we were up 10-6. Then It just kind of went to **** and 7-9-10, 26-31 was the result)

That got me thinking, because I've tweaked my builds around and played differently... but I can't tell if it's making a difference. I play Nasus mostly (Tank and AP) and I'm competent with Sivir (about 50/50 in matches with her).

The best way to improve is to focus on one thing.

ADC: Focus on getting more CS than last game at X time. Then, focus on getting that same CS while getting in harass/dodging theirs.

Tank: Focus on knowing where their champions are and knowing when to engage/peel.

And so on. Other than minute build differences, you can't really improve by just changing build items.

Also, 50/50 WL ratio is 1200 ELO. You should have a 50/50 shot of winning a game, not just going 50/50 in wins and losses.

What I meant was LoL does a reasonable job of putting you up against similarly-skilled players.

So is the fact that I'll have a winning streak followed by a losing streak a sign that I'm not really improving?

There are many areas of which you can improve upon. My suggestion is that you don't do with playing only one champion. Improving can also mean gaining more knowledge about items as well as the different champions out there.

General tips for playing well is to always move around, position yourself as safely as possible in laning phase, as well as keeping an eye on what is happening elsewhere in the game at all times (Look at minimap, and when you're back in base or something like that, try to understand how the game is going - who has the upper hand, and see if you can do something to either improve your team's lead, or to get your team into the lead, somewhat. This can be done through ganking other lanes, stealing enemy jungle, warding efficiently, and so on. Remember that you can use the minimap to adjust how aggressive you can be in lane. For instance, if the enemy jungler is ganking your bot lane, and you're in top, you know you're safe, meaning you can attack the enemy top champion if you think you can beat them. If you have Teleport with Nasus, then keeping an eye on the rest of the map is even more important, since you will want to be able to be there if it is needed, and of course evaluate if you can make a difference (You may not be able to do much if both the enemy jungler and mid are ganking bot and your mid laner isn't there - that'll just mean 3v4 which isn't super much better, but of course slightly better, and you might net a kill if the enemy is towerdiving.)

You can easily keep track of your improvement rate by a few factors.

1) Watch to see if your CS goes up game by game. Top players will have 160+ CS by 20 minutes (minus supports of course), so if you watch your CS go from 50, to 60, to 70, by that time, you're probably improving on that aspect.
2) Keep track of your lane match ups. If you started out always losing to 'x' champion, but after playing against him multiple times find yourself coming to a draw or winning, you are improving by alot.
3) Keep track of how often you change the course of the game. Whether you carry the entire team on your hulk-like shoulders of awesomeness, or are able to play General and give orders that lead your team to victory. Either way, a win is a win that you made possible. If it starts off at 1/10 games and moves up to 5/10 games, youre definitely showing strong signs of game comprehension and situational awareness.

240 cs spawn by 20 minutes, if you hit 160 then you'd have about 66%. Most top players are hitting between 85-90% cs by that point

Well, the wheels kind of fell off my improvement streak. Now I can't win any games. And I'm not joking, the only game I've won in the past 6 was a premade team.

Play a champion I'm good with? Defeat.
Play a champion that fills the need in the team? Defeat.

I'm stuck in a permanent tailspin and absolutely nothing I do can stop it. I'm really getting discouraged from all the "go back to bots, noob" and "thanks for costing us the game, loser" I'm getting after matches.

Everyone is quick to kick you, but no one wants to give advice. 3 victories in 10 games, I'm doing something drastically wrong but have no clue what it is.

I want to keep playing, but knowing I'm going to lose every match is getting harder to swallow.
