Posted a Comment: "yes, the combination is entirely random. However, combining only two runes will only give a rune of the same tier (eg. 2 tier 2 runes can only make 1 random tier 2 rune), while combining 5 runes will make one of a higher tier ( 5 tier 2 runes makes 1 random tier 3 rune)."
