I main mid so i think the only things u really need is (well otp is actually recommended the most) 1. AP champ. Having an AD champ is really good, but not completely needed depending on the meta, rn its mainly ad jngler meta. Having a champ to deal with tankier opponents is good too (ie. cassio, swain, etc.), engage champ is nice but not needed. I definately dont think assassin is needed due to the difference in play style to things like mages. If enemies is squishies, there are many burst mages/artilarry mages that deal with them effectively. Priority/Roamer is nice but not needed either.
I do like your who beats who for mid, how i think of it is midranged mages/burst mages beats assassins, artilarry mages beat midrange mages/burst mages, assassins beat artilary mages. Ofc there are lots of exceptiosn but midrange mages deal with assassins by having good dmg, still bullying with range, cc thingies, more defensive tools. Artilary mages just wave clear and outpoke/scale midranged mages. Assassins get ontop of artilary mages. Ofc there are marksman (trist, lucian, akshan, some offmeta like xayah, twitch), bruisers (irelia, sylas, diana kinda, pantheon kinda), tanks/juggernaunts (galio and random top laners that people use to destroy assassins), enchanters (soraka, karma, etc.)
Thoughts for other roles, Top i think u need an engage/tanky and/or someone to deal with splitpushers somehow. Need an Ad champ.
Jngl, ideally someone who can gank well, an ap/ad
Bot you can otp imo, ideally someone that can scale (ie. playing champs like draven/lucian with a support that does nothing isnt optimal).
Sup having a frontline/engage is nice, but ive played like 200 games of support with basically only soraka and its probably my highest wr.
These are assuming you want to cover everything, like I said before, OTP is usually recommended against players to climb the best.

I'll refine the wording for the champion pool size, I think I have the same idea as what you're saying.
I'll also take into consideration your points on the roles for each position, definitely something I need to work on a little more.
My MMR is heavily messed up since I'm getting 11 LP with a win and losing 23 on a loss, so it's pretty difficult for me to climb.
Hope more people can actually look at this guide, a lot about it I'm unsure of, hence the title Feedback Wanted :)
