I have seen a number of builds so far, and most seem to use Madred's or Trinity. My opinion on sheen is that Q is already so high burst that you can get damage and utility elsewhere. This is my take on Vayne.
The runes are a mixture of defense, offense, and utility. Full HP/level seals, Arp Reds, and CDR Glyphs. Quintessence of Health or Desolation are preferential.
This keeps you doing damage early, keeps you alive early, and gives you cdr early. Just like katarina, early dominance helps you go for that first blood!

I've been wavering between my full offensive masteries and full defensive masteries. Vayne seems to get hurt badly late game, providing the opponent knows how to focus a dps down. I've seen SilentiaReyve describe this as "Shaco Syndrome."
It may be wise to go defensive masteries, but there is something to be said for 15% increased damage and extra armor penetration.

This is where the meat is. Most builds I have seen have you go Madred's Bloodrazor or Trinity Force. Biggest problem with Madred's? 3800 Gold. It synergizes well with Silver Bolts, but you can get damage elsewhere. There are two problems with Trinity Force:
(1) Sheen synergizes well with q, but AP is a wasted stat and has other better options. A B.F. sword early adds less. but similar, damage to q, provides an early gateway into Infinity edge, and adds overall autoattack damage.
(2) Phage and Zeal can be made into more useful items, Phantom Dancer and Frozen Mallet, both of which have better utility for this character, who suffers from low HP, absolutely skyrockets with enhanced chasing and kiting. Part of this, of course, is that Frozen Mallet has a 100% chance to slow, where Trinity has a 25%.
The core struggle of this build is what do do WITH the B.F Sword. Bloodthirster adds vital lifesteal, but Infinity Edge yields Crit. Depending on how you play, specifically regarding risks and durability needed, either build would work.
In some instances I have been trading Philosopher's Stone for Cloak and Dagger, and Boots of Swiftness for Berserker's Greaves. The extra attack speed and crit enhance damage late game.
ON build two, which lacks armor penetration items, you may consider dropping dancer for cleaver (still get attack speed, more dmg, and pen) as opposed to last whisper, as attack speed is key.
