I have played a fair amount of champions and found almost all have been fairly compatible with them, about
20 champions. I just want to know how familiar should i get with a champion to use it in a rank game or some serious 1v1?

My rule of thumb was to be familiar enough to get a perfect score in a bit game.

My new rule of thumb is to be able to snowball hard enough to 1v3 in Twisted Treeline.

Just play until you're comfortable with the champion. It's up to you decide.
