They do work together, you're just using them in different spots. There are 4 kinds of runes: Glyphs, Marks, Seals, and Quintessences. Having the same type of rune in any spot will add up to a cumulative effect. For example, if you get 9x Attack Speed runes in marks, seals, glyphs, and 3x in quints (there are only 3 quint spots) you get I *think* 25% increased attack speed. Im not for totally sure, since i've only had one person ive ever met think that particular runepage was useful for any champ, (it was

Runes always stack additively. If one rune gives you X amount of stat, two runes gives you 2X of stat. It always works like that. If you have 9 marks that give X amount of stat, you get 9X of that stat, and if you have 3 quints that give Y amount of stat, you get 3Y of that stat. Then they just add together to be 9X + 3Y of the stat.

From what I know, the reason why you don't do 9 marks of magic pen and 9 glyphs of magic pen is because the glyphs don't give you enough magic pen to be worth swapping out the standard magic resist runes. There are some champs where it's better to swap out the standard MR glyphs and armor selas, but not in most cases. you can't make use of all that magic penetration if you die before you can use it.

Used both sets got any other effects, they do not work together or it do not matter?
