It pretty much doesn't matter, masteries don't really change whether an item build is bad or good at all. There's some particular types of build, such as building a melee champion with no defence, that work better at low levels. In general you can 'get away' with building stuff that would be substandard against more experienced players.
Most builds consist of a base set of items that are really good for that champion due to whatever their skills scale off, and that doesn't change with masteries or runes. After the core is built, its the same at high and low lvls, it depends on how you and your team is doing, what is killing you and how to counter it, or if nothing is killing you, how to keep the streak going.
All builds are pointers, and will always need to be changed slightly or alot depending on your farm, your team, their team and whoever you are laning against. The good guides give lots of good pointers on how to counter different kinds of situations.

Not sure if this is the right place for this but..
When browsing through the builds, as a new player, are they even effective in the lower levels (without all the Mastery points etc..) Or should I just play around with variations of the same build to see what works better at the lower levels? (playing for about 4 days now, I just hit 10)
