No. Read the description of the combiner. 2 of same quality input -> random run of same quality (IE. T2 or 410 IP T3 rune), but 5 of same quality rune gives you random run of better quality (IE. 5 T2 -> 1 T3, or 5 T3 410 -> 1 T3 820 or Quint)

Dont use combiner on T3 runes i think, save them, maybe another time you will need them.
Combining 1 x mark of alacrity( % att speed)+2 x mark of strenght ( phisical damage) + 2 x mark of insight ( magic penetration ) tier 2, you get Greater mark of strenght(+1,7 damage) tier 3...
