Won't work with LoL in any case. LoL only supports 4 mouse buttons. I know since I have 5 and can't bind the fifth one.
There is a TON of games that only handles 3-5 mouse buttons and the mouse-wheel is included in that so any more than 2 thumb buttons is rarely even usable and this mouse has 6...
The buttons also seem to be a little too far back to me. They seem like they might get in the way for comfort.

The big MOBA games don't support even close to 6 thumb buttons.
LoL supports 1 thumb button and DotA 2 will probably support 2 thumb buttons (because it's in the source engine).
So you'll basically have 4 or 5 useless buttons at the side of your mouse.. (wooooooooo...)

Surely Q, W, E, R, D and F are 6 buttons? Possibly more for A and S? Then for laugh!
