I'm used to Vent from playing other games. I prefer it over anything other VoIP out there. The only complaint anyone could have about it is the fact that setup can be quite complicated for someone who doesn't know much about it.
As of late, the group I've been playing with has been using a VoIP program called Mumble. It's like user-friendly version of Ventrillo. It uses setup wizards to guide people alone on setting up their microphone and speakers/headset. It's also nice because there are quite a large number of people just hosting out places for free and it's fairly simple to get find a host to leech off.

The biggest plus to Vent is that it can run on a dedicated server. This means less voice lag and less call drops, especially when playing with people that are far away from each other. Also Vent has a lot of extra features intended specifically for gamers. Skype is simpler to get going though, simply because you don't need a server, so in a pinch it might be a better quick fix.
I agree. I find the current version of Skype is very resource heavy which can lead to a slow down in performance of LoL. Since we all know LoL is not competitive in anyway that shouldn't matter....HAHAHAHA...
Anyway, I advocate the use of Vent until Skype gets patched because with Skype its free.
