For positioning, it's always one of the hardest part to learn, and I've included a couple of mid-late game videos from which you can get an idea. I'll see if writing a section for it is manageable, but maybe a video is the best tool. Some time is required, anyway.
1/ I think your items section in your cheat sheet is a little messy. By shortening the 2nd title, you can then fit the Core Items in the first row. As for Situational/Unusual, think about how you could break those down into "at a glance I should by" sections. Personally, I would focus on either mid or support, not both. By having the Support Start at the bottom, you've confusing people a little. Is it a support guide? Is it a mid guide?
2/ In your Threats section, you mention a lot of "you can shut x down early". I don't read many threat sections, but it seems like a lot of your comments are about having to beat the enemy early, which is sort of... obvious? Maybe think about some tips that are super useful to know. Like ie, Ziggs can use his ult to assist other lanes, so be sure to call miss after level 6... I mean, that doesn't pertain necessarily to Brand, but stuff like that. Interactions that you've discovered from playing against those champions. "You should be ok" doesn't really help a reader.
3/ Paragraph breaks/sentences. Try and keep your paragraphs "together" and double enter space between paragraphs.
4/ I would say remove the spoilers and let the gifs shine! :)
5/ If you want your guide to be super cohesive, stick to one-three colours that are complementary or in the same scheme. For example, if you're working with Brand, pick a skin you'd like to use for the entire guide and base the colours off that skin. Or if you like showing off multiple skins, try and keep the colour scheme relevant for the skin you're showing off.
6/ I like your explanation of mana worth! Though I think ultimatives are a little difficult to uphold, as there are so many "what ifs". You say "minions > poke", but is it worth it to get a minion if you get poked yourself and trade nothing back? Because after a few minion > poke exchanges, you're too low to stay to farm minions, meaning you need to recall and lose them.
7/ The game settings chapter should probably go at the end of the guide, because while it's relevant to gameplay, it feels sort of odd smack bang in the middle. Thoughts?

I've realized that for some champs I simply don't know what to add. There isn't any strange or particular mechanics you should be aware of. Do I have to keep thinking or it's ok if some of them are just "don't worry and play your game"?
That support item used to be viable some time ago, maybe I can just kill it

4. Are you sure? You're not the first one that ask me to do that, but I don't want to have a super-heavy first loading... maybe I am still using the early '00 mindset about internet speed
5. I would like to stick to red/orange/?, but then there are stuff like mana and runes that really call for a different colour. That's why for example I used a blue skin in the mana section. Do you think I shouldn't care about the topic and just go straight with red? In the ability section, I used red to evidence

Do you think I should aim to create a personalized table of contents? Or is the default one good enough if I don't have any artistic skill?

Making a custom ToC doesn't require any artistic skill since it's just BBCode. Making the banners on the other hand, is that what you meant? In all honesty, simple and clean banners don't require almost any skill and they always make the guides so much nicer. I recommend downloading the free photo editing program Photo Scape. It's not as advanced as photoshop, naturally, but it has all the tools needed to make perfectly good banners for your guide(s)!
All you really gotta know is what to crop the image to; 800x300 pixels for banners and 800x100 for chapter banners (though you change the height to whatever). And where to find nice fonts (https://www.dafont.com/). Then with the program you can play around with different filters, sharpening, blurring etc if you want to. If you don't want to, just add a nice border and some text and there you go. I whipped this up in Photo Scape in 2 minutes for example.
