Welcome to Prototype: Omnistone tierlist,
Here you will learn which champions can get the most value out of it and which don't.
Note that this tierlist is 80% just mathematics. Every champions gets 1, 0.5 or 0 points for each rune that Omnistone offers depending on their synergy with that respective rune. (EX. Sona can do really well with Guardian = 1 point, she can do decently well with Arcane Comet = 0.5 point, she doesn't do well with Electrocute = 0 points)
GREAT SYNERGY = 1 point
OK SNYERGY = 0.5 point
LITTLE TO NO SYNERGY = 0 point
At the end of point collection I give each champion +0, +1, +2 or +3 additional points, depending on how much each champion loses by not selecting a different keystone rune (EX. Kennen's main rune is out of his kit a little so he gets +3 points, Ezreal loses quite some DPS by not having Lethal Tempo but it still sin't that necessary rune so he gets +2 points, Malzahar's Arcane Comet is great for poking but it still is quite expensive to proc and his range is small so +1 point, Aatrox loses a LOT of DPS by not selecting Conqueror so he gets +0 points.)
This tierlist is also purely alphabetical, starting with Aatrox and ending with Zyra, new champions will be added alphabetically.
RESULT: Each champion can get from 0 to 17 points
As this tierlist will eventually be very long, there will be a lot of missteps from me. That's why I also post an exact reasoning behind my decisionmaking. If you think that some ranking was too low/too high, feel free to make a comment and I will recheck If I was mentally stable while writing this. Have a nice day and thank you for visiting :)

Eventually I will sure get to Kayle and even Twisted Fate as well. I already think they deserve one of the highest ratings on this tierlist so it is just a matter of time for them to appear here :)
I've been testing recently and I really feel weird about it. It sometimes feels like not having the right thing at the right moment instead of the opposite. Heard that it has a smart system to help get the right runes dependent on situations but it really wasn't like that. I will continue testing it but I don't think I get to like that much. It will be probably removed like klepto.
1) Your champion HAS TO go well with 90% of all runes. If your champion has just super hard time procing Aftershock like Soraka, Ekko or Ashe, there is just that random chance that you will have no rune for 40 seconds straight and trust me that that's a huge deal. Then the same goes to other hard to proc runes like Press the Attack or Phase Rush. Some champions just have no other choice then ignoring those runes entirely for those 40 seconds.
2) Your champion HAS TO have versatile play path. If your champion is way too late game based like Vayne or Veigar and you are in unfavorable matchup you can't possibly ignore good ol' survive runes like Fleet Footwork and go into trades with Electrocute. Same goes for early game champions like Miss Fortune when playing against late game champions. You can't ignore your big hitters like Press the Attack and get some random slow runes instead - you need to get the most of your early game otherwise the enemy ADC just catches the opportunity and outscales you easily. Prototype: Omnistone often times just directly counters your very own playstyle and that just sucks.
3) Value of this rune is still very low. Value is the only reason you would ever want to play this rune for. As Twisted Fate, you can easily proc all available runes and your playstyle isn't all that setted up anyway. But still, the value from this rune is pretty much the same as If you would pick your main reliable rune instead. That's a big problem. This rune is just way too weak and their intelligent system doesn't cover that well enough.

If you would be able to get the insane value out of many different runes except for Predator - then it would be super good that Predator can't get high rolled. That's because having an average rune which Predator is between many super good runes would be bad as Predator would put the average value of Prototype: Omnistone down significantly. However Cassiopeia is not able to get that insane value from most of the runes, making Predator a slightly above performing rune in the disposal.
Think about it like this:
0, 0, 0, 0.5, 0.5, 0, 0
Here imagine that the red number is Predator. You can clearly tell that it is one of the best high rolls from Prototype: Omnistone making it a great rune you can get from the lottery
Now If we will remove that red number from the list because Predator can't be selected on Cassiopeia, we get:
0, 0, 0, 0.5, 0, 0
The question now is: Did it make Cassiopeia better? The answer here is no, not at all. That's because even though Predator wasn't the 1 rating super good rune - it still was an above the average performing rune available. Now that it is down, there is only 1 good rune available instead of 2.
However If we invert the situation and we will pretend that Predator wasn't one of the best runes on her in for example situation like this:
1, 1, 1, 0.5, 0.5, 1, 1
Now the red number ( Predator) is a below average performing rune that only lowers the chances you get a super duper 1 rating rune. So If we take it down...
1, 1, 1, 0.5, 1, 1
...we now get much better pool of rune choices. Predator was only keeping the average value of Prototype: Omnistone down but now because it is gone, we now have higher chances of actually getting good choices.
Cassiopeia was a champion very close to the middle of those 2 occurrences but the Predator was still one of the best runes available so I rather took the rating down by 0.5 point.
