The reason i take boots and pots at the start of the match is because Vlad can harras really well with just his runes and masteries to support him and its always better to have that extra more speed than your enemy to harass them well. I usually wait till level 6 before i start initiating deffinant kills, making sure they are always close to half health is the key, if u can manage that then getting kills will be much easier, once u have your ult and you have harassed your enemy to half HP you then ghost in and use what ever it takes to kill the enemy. I usually wait till about level 9 then i start to roam the map taking ganks where they are needed.

The reason i go with this mastery set up is because not only does it boost up your AP by 5% but it also boosts your deffence by 5% so its pretty much just like maxin out the offensive masteries but better, you have that extra bit of deffence at the start of the match to harrass alot better.

the reason i max out ToB before sanguine pool is because pool is mostly used as a way to get out of sticky situations, for example turret, you would ghost in go all out on ur enemy then leave the dots to finish him off, then pool away, plus ToB is a nice harrassing technique when used along with transfusion.
