Q-W-E-R makes sense to me because that's how they're laid out on the skill bar. Plus, binding "A" to your summoner spell 1 means that you can't A+move if you want to kite (although shift+right click accomplishes the same goal). Also, if you're setting 1 to camera lock toggle, how are you activating your item in the 1 slot?
Another issue is that toggling camera lock as a way to center your champion is slower than simply pressing a key to center the screen on yourself, which could potentially mean life or death in some scenarios. I prefer keeping spacebar for screen-centering, as it just feels more natural.
Finally, having the R key so close to Q, W, and E can be very noticeable for champions with very fast combos. Ryze, for example, benefits from having those four ability keys very close together in order to execute his combo as quickly as possible. LeBlanc is another example of a champion who needs to combo her ult with her other skills in extremely rapid succession.

If you are used to playing shooter games, I imaging spacebar would feel more natural to you.
But speaking from my own opinion, and I imagine a lot of other long time league players too, the normal key bindings are the most effective.
I like Q/W/E/R because then I am close to my items, closer to my summoner spells and can make quickly decisions as a reaction when things don't go to plan, instead of having to move my fingers all over the keyboard to different postions I am ready for anything with what is already the key bindings.

I typically put my boots in the 1 key slot then put pots and others beside them.
As for combos, I don't usually use combo reliant champions but even if I did my thumb is usually near the space bar anyways.
1,2,3 slots are for activated items

A - Summoner Spell 1
D - Summoner Spell 2
(sometimes) Spacebar - Ultimate.
1 - Camera Lock Toggle
I have the other keys layed out this way because it feels more natural to go back to the traditional WASD setup like in a first person shooter or whatever else has it.
When I have my spacebar assigned to ultimate it is always a non targeted spell like Soraka's or Tryndamere's ultimate. I call my spacebar the "Oh, ****! button" because I often find myself mashing it when I get into a tight situation and start saying "Oh, ****!" It's often much faster to slap the spacebar then to tap R.
If you think you can break your hands out of the traditional layout I highly suggest you give this layout a try, especially if you play other games that use the WASD layout.
