The rune mixer is ideal when you have a lot of tier 1/ tier 2 runes (or just a lot of ****py tier 3 runes)
Because if you mix 5 minor runes, you will get a new one in the next tier, with tier 3 as the maximum.
And sometimes, you get lucky, and get exactly the rune you need, preventing you to waste IP.
If you just combine 2 runes, they will remain in the same tier, so basically for runes you won with the rune mixer, and are pretty useless, or runes that you thought were awesome, but were pretty disappointing.
Combining runes do NOT allow you to combine the stats, but will only grant you a different rune.
This ONLY works with chance, not by pick.

No. save up until you get to 20, dont buy runes before then, they are a waste.

The Combinator is nice!
Combining old Runes is really helpful! After getting access to a new Tier-Set your old Runes are just useless. You cant get IP for it like with Champions (3*Refund) and you cant use them, because they are nearly worthless.
The 5*Runes-Option:
Minor Tier Runes are only for Filling the Holes in your Runeset or to have a little luck with the Combinator to get what you want. Of course, use only the 5*Option for a higher Tier.
And if you have no Minor Runes to create a new Rune, dont spend IP for a gamble, just buy what you want.
The 2*Runes-Option:
Using the 2*Option in the Combinator is not useful. Even if you have a Tier 3 (AP) Set and want to play AD instead of AP, just buy those AD Runes. Gambling is not an Option, there are to many Runes, your chance to get what you want is to little. And you may think about playing as an AP in the future again. Using the 2* option is a Waste of IP, because you destroy Runes.
Using Runes before Lvl 20 is a Waste of IP?:
Why spending IP, the Runes are ****, compared to the Runes you can have later?
A Simple Aspect. You are stronger. If you play only bot-matches, it doesnt matter, dont buy any Runes until lvl20 and access to Tier3.
But if you like to play normal matches, you can get a high advantage. Think about it, you get 150 IP for a Firstwin. Those Minor Runes are quite cheap. An Example for ADs:
30IP
+0.94% attack speed
15IP
+0.54 attack damage
15 IP
+0.83 magic resist
15 IP
+0.78 armor
If you win more Games, because a minor Advantage, you will get those IP back soon.
Waiting til Lvl10 for Tier 2 Runes is an Option, though, you will get there fast.
I myself bought Tier1 Runes and played til, Lvl 20 without new Runes. Everything is possible without Runes, but if you play against a opponents, which matches your strenght, +2 Armor may Save your Live or get you a Kill, and maybe a Win.
