From my personal experiences, getting Q first is the most optimal, then maxing that and Lucent Singularity (E), and getting R whenever I can. I only ever got more W if my lane mate would happen to be more squishy than I (Teemo >.>). If thats the case, then its this order (maxed)

if not, I go Q>E>R>Wm maxing out my bubble last since I play a more aggressive Lux than most. Also, this order allows for better farming, better harassment and faster kills. Though you will get very mana hungry early, it is worth it in the end

I always level up my E first, the damage is high, it has a gigantic range, can be used to scout brushes, can be used as a slow. All in all it's just so useful in so many ways. Second I max up is Q, even at early game without your ultimate your Q and E can do butloads of damage.
The W is pretty good too, but imo it only starts to shine late game, make sure you hit your champs twice with 1 W.
And well your ultimate, who doesn't like gigantic lazorbeams with a 24 second CD doing around 1000 damage.
