In the case of Illumination (which as Luther pointed out, no one has calculated in), Lux gets a small bonus from mPen and not AP quints unlike most AP mids. According to lifebaka, her ultimate and early-mid game damage will rise a little with mPen assuming the enemy sticks with their base 30 MR. A level 6 Lux with that hits an enemy laner with a full combo including passive procs should (if I understand your math correctly) get a bit more damage out of it than one with 15 more AP instead.
Considering the early game advantages and the likelihood of an enemy AP having more than 30 MR, I think it's safe to say that mPen quints are not so optimal for most APs, especially those with high scalings. They could probably work on Lux in certain scenarios, but in most cases it's simpler to just stick with the AP quints as they're less dependent on the enemy MR and stage of the game.
In my own games I will probably be using MS quints on Lux since I get a lot of my damage from passive procs and autoattacks early game anyway. It's mid and late game that I'd rather rune for, so MS seems to be the best option in my eyes.

I think Magic pen quints are fun to roll with though.
It's really funny if you're playing Kassadin for example
and you're laning against an Anivia and you use your Q
