If I am playing Singed and am constantly casting poison trail would that could as one spell being cast or would it just check every 3 secs to see if I am spending mana?
Just wondering if I need to turn the trail off and back on to get the extra mp's.
If you leave Singed's poison trail toggled on, you will get bonus mana every 3 seconds.

You don't have to hit anything with it, just cast a spell (/be spending mana now). If you're playing a champ with any skill shots, you can shoot them while you're in the base, or headed back down the lane, and get some extra mana that way. Just be spending mana as often as possible if you're mostly focused on building it up.

Also to others it was changed to whenever you spend mana so now it does work with Singed.
I just checked. While on Anivia, get your Tear/AA when you already have Glacial Storm. Just sit on base and activate R. You'll see the mana bonus grow in steps of 4, uninterrupted. If your team can afford 188 sec of you doing nothing, and I'm against it, you'll have capped the 750 bonus. If you rush AA skipping Tear, you'll have Seraph's after 188 sec. For a better use of time, go spam your mana on minions and champs. Or better, just play along and have fun.
Then they've since changed the way the item works. Back when I used to get it (pre-Seraph's Embrace, season 2, and when it used to give +25 mana regen), you had to cast a spell, not just be spending mana. That's good to know, though. Makes it even easier to increase the bonus mana with Anivia.
As for why you aren't noticing an increase in max mana, Seraph's Embrace doesn't give you any more mana than a fully stacked AAS. They just change the amount of mana it gives you in the base stats, and remove the bonus mana part of the item.
I've never played Singed, so I'm not sure how his spells operate, however, if it's a toggle, both the on and off "casts" count as casting a spell. The item has a CD of 3 secs, and you can gain bonus mana every three seconds. Toggling spells such as

My Anivia was about ~2800 mana when AA Staff was almost at 750 cap mana bonus.
The instant AA turned into Seraph's, the new item shows 1000 bonus, but this bonus didn't appear in my stats.
It seems that the logical path is Tear -> +250 + 750 (fully capped-charged)total bonus 1000, AA -> +250 + 750 (the bonus is still 3sec-ticking if you didn't reach 750 cap yet) total bonus 1000, -> Seraph's 1000 total. It may happen that you already have the total 1000 mana bonus of Seraph's if you already capped Tear's bonus. The difference between the Tear->AA->Seraph's will be the other stats besides mana. Telling this for those guys, like me :-\, thought on have a huge instant leap of 1000 extra mana.
