A personal favorite - the caitlyn challenge is the first thing that I do to teach my students have to balance farm and lane presence, and pressure . You get a high ELO player to 1v1 you in the bot lane. You pick some ADC other than caitlyn, and he picks caitlyn.
You'll lose lane, but you'll learn a lot about how to position in lane to get the optional balance of lane presence, and safety.

I would say the only challenges you should set for yourself are ones you have control over, such as cs @ 10 or 20 min and proper vision control around your lane if you're a solo laner (though I suppose it's a bit harder to measure what "proper" vision control is).
Setting goals in terms of kills and assists is a lot trickier / more unrealistic and can lead to playing too aggressive in situations in which require a more subtle approach. A better alternative to this would be to set goals in terms of kill participation.
The same goes for deaths: while it is generally good to aim for a low number of deaths, it is always more important to do the right thing (a.k.a. ulting 4 people as
Malphite so your team can win the fight and then dying > not ulting in the same situation because you promised yourself not to die more than the 3 times you have died already).
