I can confirm Karthus is a damn good pick for a defender, simply put him at the top-mid point or your team's bot point and have him stand there all game. If he gets attacked, even by two or three guys, he can usually take several down with him, or at least delay them for a long time until backup can arrive.
Because of his passive you can kill them if they try to cap, or simply delay them from capping. It also makes his death timer like one-two seconds near the start of the game so you can run back almost straight away.

Highlander + Ghost + Youmuu's Ghostblade + Priscilla's Blessing = You can't be caught, no one can escape you.
You'll be dealing TONS OF DAMAGE.
The great thing about Dominion is it's a 5v5 roamer's heaven. Enemies are typically separated. So Master Yi has numerous chances to kill and snowball with Highlander.
