Start playing some co-op vs. AI to get you back in the motion. After a few games of that, if you feel you're ready, you could start playing regulars. Try to read guides on the champion you plan to use though, they might re-introduce you to the new mechanics of Season 3.

spend some time looking at the new items. Some newer items such as spirit of the Elder Lizard, or Iceborn Gauntlet have become a huge part of the game, and a lot of items build out of newer items, like Zhonyas now builds out of Seekers Armguard, a great item to pick up vs melee casters in that mid lane.

Take all the advices given by the other about starting co-op or normals to get yourself familiar with the game.
but if you looking to get competitive again in tanked, I suggest learning 1-2 roles and 2-3 strong champion in each. Examples proceed (mind that it greatly depends on your playstyle):
Top : Jayce, Jax, zed, renekton, kennen and cho'gath (safe pick)
Mid: Ryze, wukong, ahri and kha'zhix
Ad carry: a lot of viable champion but the strong ones remain: ezreal, cait and miss fortune (game changing ultimate for solo q)
These are base on my main roles and opinions so don't take them word for word.
Season 3 is much more based on mobile champion such as assassins (not so much any more due to the recent nerf but they are still strong).
Try to avoid learning slow squishy champions: e.g veiger (can get easily countered due to ad casters occupying mid), heimerdinger (getting reworked soon) and urgot (only works effectively in some comps.

If you've played before you just need to get updated on curent meta and new items and you're good to go. Play normals (or co-op but I hate those) and spam whatever you liked playing to get the kinks out and then get serious.
