Leona + Tristana, but I'm more interested in knowing when to do them then when not to :P

Also, do Zilean and Karma have any optimal bot lane combo or is it just that agreeable concept of "send them mid"?

They are really strong early against most combos but imo they excel against low mobility champions like Ashe, Miss Fortune, Varus with either Sona or Soraka support.
I wouldn't recommend using them against Alistar.

I never see them as support in ranked tbh, so idk.

I caught a Karma/ Graves combo once on my side, they completely facerolled botlane (Soraka + someone with low mobility, not sure), their coordination was kinda unnexpected for my ELO. Karma really knew what she was doing, was quite impressive, but I understand that the girl still needs some sort of improvement in order to go bot lane

Amazing burst combo, but I donno, Karma kinda fell lackluster without farm.

Sivir + Soraka is a mean combo, massive spam with Boomerang Blade and high lane control.

Sivir/ Soraka combo is my dearest favorite. This setup can farm an entire minion wave starting as low as level 4, clearing the wave extremely quickly and efficiently while Soraka provides unlimited mana and healing.

I've seen a really aggressive Darius + Draven bot lane take it home, both with ignite. Dot too damn stronk.

Harass < sustain <kill lane< harass

Kill lanes can't go for kills cuz they'll get harassed and have a disadvantage. The rest is obvious.
