It can be sometimes actually. Without an opponent last-hitting too, your minions become more numerous than the enemy's, and several times you're going to have your entire minion wave of 8+ minions all attacking the same enemy minion. It can be quite hard to time your last-hit on it when that happens early-game.
But yeah, that's not very helpful. The only reason you'd be doing bad in cs in a game isn't because you're bad at last hitting, it would be because your enemy is harassing you and you can't risk getting every last hit because you lose too much HP. You've just got to try and harass them back, and if you end up lower HP than them, then it seems you have an unfavourable match-up, unfortunately, by which point you should ask your jungler to gank a few times.
This happens to me as well Babit. So frustrating. My problem is that I tend to focus on a certain champ for awhile, practice, do well, and then I hit one of these ruts and just feel like all of my work on that champion has been a waste.
The best solutions I have, are either:
1. Playing an ARAM. I know it sounds stupid, but somehow knowing that I can't pick a character and that the game itself is just chaos makes me feel better. Even if I lose, I almost always enjoy myself and it is a great stress-reliever.
2. Switching to some sort of physical activity for awhile. Go run, go walk, lift some weights. You can burn off some of the frustration!

You can't peddle papooses with a dancing monkey.
^ I like the #2 option there.
Kickboxing is what does it for me.

Which is why I don't think practicing last-hitting in a custom is very useful. Maybe for newer players who have trouble just farming gold, but I think by the time you hit 30 you should be able to last-hit pretty well as long as you aren't zoned from the cs.

Thanks to Lugignaf for the sig!

I have some solutions too, but mine are a bit crazy:
1. Go get a pillow and punch it. Punch it as hard as you can.
2. I have a really old washing machine. So I take a bat and whack it as hard as I can. Relieves a lot of frustration on my part.

^ This.
There is probably some science behind it, most likely something to do with happy neurotransmitters. And when you get back you don't have to feel guilty about sitting in front of a computer for aaaaages at a time lol :P
that's what I do, or on custom games with a friend, practice last hitting on off days, just some friendly harass. Gets good after a while.
