This is Pyke's biggest counter. If Lulu saves her polymorph for whenever you engage, she can completely stop you from doing anything in team fights since the ability is point and click. I recommend banning every game you plan to pick Pyke.
Morgana
Since Morgana tends to rush Zhonya's Hourglass as her first-second item, she can save it to counter your ultimate. Her Q and ultimate are also very good at stopping Pyke from engaging as the stuns and roots last 3 seconds (without tenacity). Morgana's E also saves her and her teammates from any chance of Pyke hooking or stunning them during fights.
Zyra
Zyra has good poke in laning phase against Pyke, allowing her to widdle him down and kill him with a good E+Ignite combo paired with her ADC. She can also build Zhonya's Hourglass to negate Pyke's ultimate execute and reset. Despite this, she has little to no mobility and is extremely squishy. If she misses her E pre level 6, use that time to engage on her as she lost all of her CC and has no way of running away without using a summoner spell or being saved by her team.
Yuumi
Yuumi isn't much of a threat to Pyke as she is just annoying to play against as Pyke. Since she can't be hooked or stunned while on her allies, Pyke loses out on a lot of potential kill power on her and the ally she's attached to due to her passive giving them a shield and her E being able to heal them. Her ultimate is also really good against Pyke. The reason she's a minor threat, though, is because laning phase is very difficult for Yuumi when she wants to get her passive. If you see Yuumi walking up to auto attack you for a shield, engage on her. If she hops to her ally immediately, turn on her ally. Yuumi is also extremely squishy, has no mobility, and rarely builds Flash or Zhonya's Hourglass, allowing Pyke to chase her, one shot her, and get an easy reset on his ultimate when she is alone. One other thing that makes Yuumi pretty weak against Pyke is the lack of vision she can place. Since she's so squishy, she relies on allies to bring her everywhere she goes, meaning she can only ward where her allies go. Use this as an opportunity to sit in a bush and take her and her allies out when you know they're going to try warding.
Nami
Nami's Q can stop Pyke in his tracks while he is trying to hook. Since Pyke loses movement speed while he is charging his Q, she can hit him easier with her Q, stopping it completely. She also has a bit of movement speed for her and her allies which make it harder for Pyke to hit his abilities.
Soraka
If Soraka saves her E until Pyke begins charging his Q, she can stop him completely with the quick silence. It also stops Pyke from using his E to engage for a short time. Soraka's ultimate can save both her and her teammates from his ultimate easily, but she has little to no mobility throughout the game, allowing for an easy kill if she's separated from her team or mispositioned.
Taric
Taric's E can stop Pyke from engaging in fights and his Q can heal him and his allies a bit which may deny Pyke kills, although the biggest threat is the invulnerability from his ultimate that completely counters Pyke's ultimate and denies him resets.
Zilean
Zilean's Q+Q combo can stop Pyke dead in his tracks when he tries to engage, and the slow on his E can slow him down further, denying a lot of engage potential. Plus the amount of poke Zilean has against a squishy champion like Pyke can cause major issues in lane. But the worst part is his ultimate which can save allies from death and deny Pyke ultimate resets.
Renata Glasc
Renata's ultimate isn't very effective on Pyke, but her Q can deny engage and help her escape and her W can save her and her allies from his ultimate, so be sure to bait it out before going all-in. Don't ult an enemy who has Renata's W unless they're the last one alive on their team (or part of the last 2 alive).
Alistar
Alistar's all-in potential and CC removal on his R makes him a big threat to Pyke in both the early and later game.
Rell
Another one of Pyke's bigger counters, Rell is able to stop his engage and completely turn fights around in the press of a button. Due to her high health and armor (if she builds it), Pyke is also unable to do much damage to her on his own. The only thing that Pyke can do is bait out her W and let his team do all the damage as she tries to waddle away.
Amumu
Amumu is pretty strong against Pyke as his Q+R combo is able to quickly deny Pyke of any mobility and engage.
Bard
Since Bard tends to roam a lot, just as Pyke does, bot lane will be a 1v1 lane around the 8 minute mark. Bard's ultimate can save him and his allies from Pyke's ultimate, but since the cast time is really low, if you have a quick enough reaction time, you shouldn't miss your ultimate to Bard's R. Just don't follow him through his portals (E) unless you know where all of his allies are and his Q is on cooldown.
Brand
Brand melts people, but can also be one shot pretty easily. Laning phase will be difficult as he can easily widdle you and your ADC down, but after laning phase, he should be easier to kill as long as you dodge his Q. Since Pyke has a lot of invisibility off of his W and Duskblade, he should be able to stop Brand from using his E and R on him, although if Pyke has already been hit by one of Brand's abilities, the enemy can see Pyke even when he is invisible.
Blitzcrank
As long as you or your ADC don't get hit by Blitzcrank's Q, laning phase should be easy, although Blitzcrank's E can completely negate Pyke's escape and engage, but can be played around if played well. Later on, Blitzcrank's R can stop Pyke from using his abilities for a moment and can cancel his Q charge if he's too close, so make sure to stay far away from him and focus on his allies in teamfights.
Braum
Braum is a very minor threat to Pyke, as his E is basically useless against Pyke. Braum is better at fighting against champions with poke such as Ezreal or Xerath, but not so much someone like Pyke. Just make sure you save your Q for the enemy ADC as Braum can dash to them whenever he likes which can allow for an easy bait and switch into his R if you miss your Q.
Fiddlesticks
As long as you go one or two tenacity runes, Fiddlesticks shouldn't be a big problem early game in lane. Since Fiddlesticks is also very squishy, he tends to rush Zhonya's Hourglass, so don't get baited by it when you decide to go in and try to execute him.
Karma
Karma is a very annoying champion to play against as Pyke. Since she has so much movement speed, shielding, and high damage poke, laning phase is nearly impossible for Pyke. Despite this, her mid-late game against Pyke isn't nearly as strong as long as you don't get targeted by her W in fights. Wait for opportunities to arise and go in when you know she doesn't have her W or R up.
Janna
Her Q and R can stop Pyke from engaging, but that's pretty much all she can do. Pre level 6, fighting Janna should be easy once her Q is down and engaging shouldn't be a problem as long as the enemy ADC doesn't have hard CC.
Leona
Leona is extremely tanky, even early game, and has a TON of CC, denying Pyke of any attempts to engage. Even if she doesn't target Pyke, it's still very difficult to kill her with her W and tanky items, making her a big threat both early and late game.
Lux
As long as you are able to dodge her Q effectively, then fighting her is a breeze, although laning phase against her is difficult due to her high damage poke and shields for both her and her ADC. Later game, she's an easy kill as long as you dodge her Q.
Maokai
Save your E for when Maokai engages on you and you can turn fights around easily. If he uses his full combo on you, just E away since his damage is so littler and Q him into your teammates if there are no minions in the way since he'll be on cooldown for a moment. Laning phase is really easy, but later game (after level 6) will become a bit more difficult due to his ultimate.
Nautilus
Nautilus is very strong against Pyke as he has a ton of cc and can deny Pyke of his escape if he decides to save his E. As long as you don't walk up too close to him, then it shouldn't be too difficult to get away unless he flash+auto attacks you, meaning you need to dash away and try to dodge his Q. If you're too far in a fight and he's level 6 or more, he can just press R on you and stop you from doing anything in a fight.
Pantheon
Pantheon has a lot of high damage in the early game and his E can block your ultimate execute, making him extremely dangerous in laning phase. Despite this, his late game is extremely weak and he'll usually fall off pretty hard (unless you're against Spearshot, but he doesn't play support so that shouldn't be an issue).
Poppy
Rakan
Senna
As long as you dodge her W, she's an easy kill due to her squishiness both early and late game. Just be aware of her attack range and don't let her poke you down by either playing safe and far away from her, or by playing super aggressive.
Seraphine
Seraphine's E and R can stop you from doing much in any fights, but since they're both skillshots and Pyke is a very mobile champion, if she misses, that's your queue to engage and kill her. If you aren't confident in your ability to dodge any of her CC abilities (and anyone else's for that matter), go tenacity runes.
Shaco
If you ignore Shaco in laning phase and team fights, he becomes useless no matter what items he builds. Just don't chase him anywhere when he hits level 6 or if he builds AP, as he can melt Pyke easily this way.
Sona
Sona's only CC ability is her ultimate. If you go tenacity runes, her stun duration is greatly decreased. Pyke destroys Sona in laning phase, especially if she gets too close. Abuse her in laning phase and get fed quickly off of her.
Swain
Since Swain tends to build Zhonya's Hourglass first or second item, he can stop Pyke from getting an execute with his ultimate. Also, Swain's ultimate gives him a lot of HP and steals the HP of you and your allies, making him pretty difficult to deal with levels 6 and up. His E is also very strong against Pyke as he can full combo him and deal a ton of damage in the blink of an eye with it. Pair this with an ADC and you'll die almost every single time he hits an E on you. Laning phase is hell against him, so roaming would be a good idea as long as him and his ADC don't hard shove the wave under your tower (which is what they should do).
Tahm Kench
Although Tahm Kench has a lot of CC with his Q slow/stun and his W knockup, he's pretty weak early game without his ultimate. Save your E when he hits level 6 so you can just dash away after he uses his R on you. His E is also very weak against Pyke as Pyke's ultimate ignores shields. If his healthbar is red, even if he has a shield that fills his health bar, you can still execute him easily. If you see him start to use his W, try to CC him out of it or away from your team when he emerges.
Thresh
Thresh can stop your engage pretty easily, but that's pretty much all he can do. Let your teammates break the walls of his R and just chase him down if you want to kill him. He has very low damage and his CC is not as strong against you as you may think. Just don't E onto him when he has his E up, as he can redirect your phantom and cause you to miss your stun easily. Don't charge your Q when you're near him, either, as his E can also stop your Q charge.
Vel'Koz
Once Vel'Koz uses his E, he doesn't have much escape against you. If he uses his E in a fight, use that as an opportunity to go in and kill him. Just be weary of the fact that although he is squishy, he can still build Zhonya's Hourglass which can stop your ultimate from executing him.
Xerath
Xerath's poke in lane paired with his high damage output with comet/first strike is pretty hard to deal with early game. Even mid-late game can be difficult since he can kill you easily after hitting his stun due to the fact that Pyke is so squishy. If you can hook him, he has no escape potential, so it should be easy to kill him if he's alone or you have your team with you.
Caitlyn
Caitlyn is probably one of Pyke's best duo laners bot lane. Pairing Caitlyn's high range and W with Pyke's hooks and stuns, they can both do a lot of damage to the enemy, even if they try to run away. If Caitlyn has traps laying around, look for an angle to pull enemies into them with your Q.
Seraphine
Seraphine's E and ultimate work very well with Pyke's hooks and stuns. Seraphine can also give Pyke movement speed with his W and stops the enemy team from using any escape tools during fights; allowing for Pyke to get resets much easier.
Ezreal
Ezreal needs to hit his skillshots to deal any damage to the enemy. Since he's also really squishy, he can't really engage with Pyke, but he can still deal damage from afar after a hook or a stun since the enemy will be standing still. Not very good, but not bad at all either.
Jhin
Pyke's Q doesn't just reel enemies in. It slows them as well; allowing for Jhin to get easy auto attacks and makes it easier for Jhin to hit his W and R. Similar to Pyke's synergy with Caitlyn, he can Q enemies into Jhin's traps, allowing for the trap to (usually) deal damage if the enemy doesn't have a way to escape.
Varus
Hitting an enemy with a Q or E as Pyke allows Varus to easily get auto attacks off, hit his Q, and even hit his ultimate WAY easier. But it's hard for Varus to engage with Pyke since he doesn't have the mobility to keep up with Pyke and can easily be one shot by the enemy.
Ashe
Ashe's auto attacks, W, and R all can help Pyke to hit his skillshots easier, allowing for a strong combo together in lane. Despite this, Ashe is immobile, making it harder for her to keep up with Pyke when he engages.
Tristana
Since Tristana can engage with Pyke and escape just as easily after scoring a takedown, they both synergize really well with each other. Since Tristana is weaker early game and way stronger late game, laning phase would usually be a lot slower. But if Pyke is paired with Tristana, he can potentially help her snowball and get fed earlier than she normally would.
Kai'Sa
Kai'Sa is pretty strong when paired with Pyke, as her passive stacks with CC. Although there are better champions to go with Kai'Sa, the fact that Pyke can give her 2 stacks of her passive on an enemy or 2 is pretty strong, especially in the early game where she has less attack speed and less chances to stack up her passive. Later game when you are engaging more, it makes it easier for her to catch up with you because of her ultimate being able to dash quickly to enemies marked by her passive, allowing for some really nice synergy both early game and late game.

Since Pyke's Q is not only a hook but also a slow, this allows for Twitch to easily catch up to enemies that Pyke hooks or stabs with it, giving Twitch some really nice setups for kills both early game and later game.

Pyke is a roaming assassin support. Able to pick up kills in other lanes to help his team get fed, he can also grant the gold from his executes straight to his allies, allowing both him and his allies to snowball. With his strong early game and high kill potential, he can become unstoppable if played right.
But he also has his drawbacks. For starters, his passive causes Pyke to lose out on health from any items that may grant health to other champions, meaning he is very squishy. His engage may also be his downfall if played incorrectly or if it has bad timings. Pyke also relies on lethality items, making it almost impossible to buy other items since he scales so much with lethality.
Pyke is easy to lose with just as he is easy to snowball with. If you can't snowball in the early game as Pyke, your mid-late game may be a bit difficult.
Combos
Pyke's bread and butter combo is a charged Q into his E. Hooking an enemy into yourself then dashing over them with E to stun is his most basic yet most effective combo. Pair this with auto attacks from Hail of Blades or the close range explosion from aftershock and you can deal a high bit of damage every 10-15 seconds early on.

Basic:
Basic Combo - Q > E > Auto
Flashy Combo - Q > E > Flash (Flash right before Phantom collides with Pyke)
Kill Combo (against squishies) - Q > E > Auto > R
Gap Closing Combo - E towards far enemy > Q > Auto
Another Gap Closing Combo - E towards far enemy > W (to make sure your E hits) > Q > Auto
Advanced:
Quick Slow Combo - Q+Flash (in that order) > Auto (with Duskblade for extra slow)
Flash Hook Combo - Q Charge > Flash > E
Multi-E Combo (Situational) - E into one enemy > Flash behind a different enemy right as phantom collides with Pyke
Stun Hook Combo - E (make sure enemy is stunned) > Q (walk back in front of them to Q them into your teammates)
Prowler's Claw Combos:
Easy Stun Combo - E > Prowler's Claw
Gap Closing Combo - Prowler's Claw > E/Q (either works)
Pyke Main Big Brain Combo - E > Tap Q+Flash > Prowler's Claw (before phantom catches up with Pyke) > Auto > R
Gameplay Tips and Tricks
Roaming is Pyke's specialty. Picking up Oracle Lens and Umbral Glaive allows Pyke to easily clear vision and pick up kills in other lanes. Once you hit your Mobility Boots power spike, don't roam right away. Wait until your lane is pushed or, if your ADC is a god and somehow got 8 kills before level 6, you can just perma leave the lane and help your other teammates get fed.
Pyke is an early-mid game champion, but his late game is very weak, so make sure to snowball and end the game as quickly as possible when the enemy team has scaling champions (Kayle, Master Yi, Kog'Maw, etc.). It will be impossible to do anything as Pyke later game when everyone has items and levels since he can die easily and is useless in team fights without a team full of CC.
Don't always go for hooks on people. I see this mistake everywhere and even I make this mistake when I get fed and crave more kills. Stick with your team as you are very weak when you are alone (except against squishy ADCs farming alone in side lanes). If you do go out on your own, make sure you have your escape tools ready at all times just in case you get caught. Do not face check bushes! This is an easy way to get CC locked and killed. Later game, this can cost your team an objective or even the game.
Peel for your carries. As Pyke, you will be getting a lot of kills due to your R resets, but don't let that get you thinking you can 1v5 the enemy team. Just because you are full build and the enemy team isn't even at their third item yet doesn't mean you can go in and kill them all alone. Stick with your team. Remember that you are a support and you can die easier than any ADC can!
If the enemy team has a ton of scaling champs (3+) then make sure you understand the risk of picking Pyke into them. If you can snowball early and take advantage of the scaling team's weak early game, you may be able to win. But if you mess around and take too long to end the game, then you'll become more useless than a caster minion. Always see what the enemy picks before locking in Pyke (and always ban Lulu).
Pyke's passive allows him to heal whenever he's not in view of the enemy after being damaged. Since his W gives camouflage, he can use it to heal up quickly while staying in lane.
Conclusion
I have been playing Pyke since he was released in 2018 and he has been one of my favorite champions to play as since then. I have a collective amount (across multiple accoutns) of around 300-400k mastery points and I've spent more time playing Pyke than any other champion. I have tried many builds on him in many lanes and I believe that lethality and support are his best bets. I love how annoying he is to play against when I play him and it always feels amazing when I get multi-kills with his R. He gets gold faster than any other champion in the game (if you can land your R) and is easily one of the strongest early-mid game champion in the game.
