Cleanse: Vayne is automatically the focus of hard cc/exhaust. Immediately cleanse these and continue wreaking havoc. Reaction time is extremely important here.
Just follow this build order and you'll be right as rain. I build zeal/PD first because vayne just scales amazingly with attack speed, with a vamp scepter thrown in I believe this start is better than bloodthirster first builds.
Build a warmogs as your defensive item, its just broken, you can also build banshee's, randuin's, or frozen mallet, all situationally of course. I usually pick up a giant's belt or catalyst as my 3rd "major" item for the mid game survivability, then finish it later.

Some people max vayne's W (silver bolts) or her E (condemn) first. However, her tumble is such a versatile ability that maxing it first is going to give you the best overall effectiveness with this champion.
Maxing silver bolts second will give you the damage you need to handle mid-late game HP stackers, or really just anybody.
Finally max her E last, it's primary use is escape/stun, therefore doesn't need points until you have too.

Ok, 21 points in offense, obviously is going to improve your damage the most possible, just take the basic physical damage masteries here.
9 points in the defensive tree. grabbing veteran's scars is just so very important, the extra hp is going to help a ton, while the 3 points in armor and 1 in magic resist are gonna help in trades and the like.
A lot of vaynes used to build her with 21/0/9 masteries. However I don't believe vayne really requires mana regeneration like some other champions might, with proper skill management and general play, I usually never run out of mana in lane as Vayne.

Attack damage quints: no secrets here, gives you the damage to deal, well damage, also last hitting
Armor pen reds: you NEED armor penetration, if you don't have it, your gonna do 0 damage, plain and simple, pretty much every bot laner + bruiser + jungler, takes armor seals.
Armor seals: no secrets here either, you need the armor to survive, overall the best benefit of any seal and I guarantee nobody will tell you otherwise. just buy them!
Magic resist/lvl glyphs: ok this is a little bit different from the average runes you see for adc. Most adc out there will be taking flat MR blues, and while that's not a bad choice (helps with harass from the support in lane is probably their only use). I take scaling resist blues because as the laning phase ends, they are going to give you a way better chance of surviving VS the enemies AP carry.
