I don't think it's ever worked like that, but I could easily be wrong.
Mystic Shot has always been sort of an auto-attack, so I don't think it would apply Rylais.

I'm 90% sure it used to slow with Rylai's, but now that you say it hasn't I'm getting a little uncertain. I'm pretty sure that it works with
Parrrley so I don't see why it wouldn't work with
Mystic Shot.
Edit: Watched a couple of Siv videos of AP Ez and it seems like I was completely wrong,
Mystic Shot does not proc spell damage.

Eh, just get
Iceborn Gauntlet if you need the slow. If you're going AP, I wouldn't worry about CC and just get a lot of damage for your ultimate and E-W burst. Try something like this:

Because Cho's spikes are not an ability that procs on-hit effects, it is an on-hit effect. Talking about on-hit effects, some proc them, some not.
The most interesting thing is that stuff like
